چکیده

Several lines of evidence, albeit most of them circumstant ia l, support the protease-antiprotease imbalance concept in the pathogenesis of emphysema. Neutrophil elastase (NE) has become the key enzyme for several reasons. NE is insufficiently inhibited by a( antitrypsin (AAT) (a 1 -proteinase inhibitor) in inherited deficiency states when the extracellular level of the predominant antiprotease AAT is low as in AATdeficiency, PiZ, and other rare genetic variants. NE .is the target enzyme of AA T, the rate of association between them being extremely rapid (1]. AAT is the archetype of the so called serpin (serine proteinase inhibitors) super family of highly homologous proteins, including the protease inhibitors a 1 -

Role of elastases in the pathogenesis of chronic obstructive pulmonary disease: Implications for treatment

Neutrophil elastase, metalloproteinases, and their inhibitors play an important role in the development of chronic obstructive pulmonary disease (COPD), resulting in extensive tissue damage and malfunctioning of the airways. Nearly fifty years after the protease-antiprotease imbalance hypothesis has been suggested for the cause of emphysema, it is still appealing, but it does not explain the co...

A human surfactant peptide-elastase inhibitor construct as a treatment for emphysema.

Two million Americans suffer from pulmonary emphysema, costing $2.5 billion/year and contributing to 100,000 deaths/year. Emphysema is thought to result from an imbalance between elastase and endogenous inhibitors of elastase, leading to tissue destruction and a loss of alveoli. Decades of research have still not resulted in an effective treatment other than stopping cigarette smoking, a highly...

Ceramides: a potential therapeutic target in pulmonary emphysema

BACKGROUND The aim of this manuscript was to characterize airway ceramide profiles in a rodent model of elastase-induced emphysema and to examine the effect of pharmacological intervention directed towards ceramide metabolism. METHODS Adult mice were anesthetized and treated with an intratracheal instillation of elastase. Lung function was measured, broncho-alveolar lavage fluid collected and...

Prevention of leucocyte elastase-induced emphysema in mice by heparin fragments.

Heparin and its derivatives inhibit human leucocyte proteinases i.e. elastase and cathepsin G, but do not inhibit porcine pancreatic elastase and Pseudomonas aeruginosa elastase. In vitro experiments, reported here, also indicate that elastin, one of the physiological substrates of human leucocyte elastase (HLE), could decrease by 30-fold the inhibitory potential of an hexadecasaccharide hepari...

IL-17A is essential to the development of elastase-induced pulmonary inflammation and emphysema in mice

BACKGROUND Pulmonary emphysema is characterized by alveolar destruction and persistent inflammation of the airways. Although IL-17A contributes to many chronic inflammatory diseases, it's role in the inflammatory response of elastase-induced emphysema remains unclear. METHODS In a model of elastase-induced pulmonary emphysema we examined the response of IL-17A-deficient mice, monitoring airwa...
